Comparing 4-AcO-DMT to Other Psychedelics



4-Acetoxy-N, N-dimethyltryptamine (4-AcO-DMT) was first synthesized by Albert Hofmann and Franz Troxler in 1963 and it remains poorly studied with regards to both its pharmacology and subjective effects.

Many users have likened the experience of taking this drug to that of using psilocin and psilocybin, two naturally-occurring tryptamine compounds found in certain species of mushroom-forming fungi commonly referred to as "magic mushrooms." 4-AcO-DMT is one of the most potent tryptamines known, with an EC50 value for 5-HT2AR at 527nM and high amplification ratios at both 5-HT1AR and 5-HT2CR receptors, suggesting it acts as a widely effective regulator of downstream signaling after binding these receptors - making it comparable to classic psychedelics such as LSD or MDMA in this respect.

Lab Testing

Users have reported similar experiences to psilocybin when taking 4-AcO-DMT, such as geometric visual patterns, time distortion, introspection and ego loss. Onset is typically 20-40 minutes with peaking between 2-3 hours and lasting up to 6 hours before feeling of euphoria, spirituality and deep meaning emerging within minutes or hours; sometimes combined with other psychoactives to enhance or prolong effects or duration.


Lab testing is essential to understanding the quality of psychedelic substances like 4-AcO-DMT.
